Description: The galactose regulon has been utilized for many decades to control gene expression with GAL1pr potentially being the most widely used inducible promoter in budding yeast1,2,3. GAL1pr is tightly repressed in the presence of glucose by the Gal80 and Mig1 repressors4. In the presence of galactose, GAL1pr is induced more than 1000-fold5. Glucose repression induces transcriptional downregulation of GAL regulatory genes. To avoid delays when switching from glucose to galactose, cells are typically grown in non-inducing and non-repressing raffinose medium.
